Quantification of Trimethylamine-N-Oxide and Trimethylamine in Fish Oils for Human Consumption.

Supplementing fish oil is one of the strategies to reduce the risk of cardiovascular disease, the leading cause of death around the world. Contradictorily, fish oil may also contain trimethylamine-N-oxide, a recently emerged risk factor for cardiovascular disease, as well as one of its precursors, trimethylamine. A method suitable for routine quantification of trimethylamine-N-oxide and trimethylamine in fish oil with a quick and easy liquid extraction without derivatization has been developed. Liquid chromatography with tandem mass spectrometry detection was employed along with a zwitterionic hydrophilic interaction liquid chromatography column and a gradient elution with eluents containing 50 mmol/L of ammonium formate. An internal standard (triethylamine) was used for quantification by mass spectrometry with an external calibration. The assay proved high linearity in the ranges of 10 to 100 ng/mL and 100 to 1000 ng/mL for trimethylamine-N-oxide and trimethylamine, respectively. The lowest limit of quantification was determined to be 100 µg/kg for trimethylamine and 10 µg/kg for trimethylamine-N-oxide, with the limit of detection at 5 µg/kg and 0.25 µg/kg, respectively. Accuracy ranged from 106-119%. Precision was below 7% the relative standard deviation for both analytes. The method was successfully applied for the determination of trimethylamine-N-oxide and trimethylamine contents in nine commercially available liquid fish oils and three commercially available fish oil capsules, showing that trimethylamine and trimethylamine-N-oxide are not present in highly refined fish oils.

An efficient strategy with a synergistic effect of hydrophilic and electrostatic interactions for simultaneous enrichment of N- and O-glycopeptides.

N- and O-glycosylation modifications of proteins are closely linked to the onset and development of many diseases and have gained widespread attention as potential targets for therapy and diagnosis. However, the low abundance and low ionization efficiency of glycopeptides as well as the high heterogeneity make glycosylation analysis challenging. Here, an enrichment strategy, using Knoevenagel copolymers modified with polydopamine-adenosine (denoted as PDA-ADE@KCP), was firstly proposed for simultaneous enrichment of N- and O-glycopeptides through the synergistic effects of hydrophilic and electrostatic interactions. The adjustable charged surface and hydrophilic properties endow the material with the capability to achieve effective enrichment of intact N- and O-glycopeptides. The experimental results exhibited excellent selectivity (1 : 5000) and sensitivity (0.1 fmol μL-1) of the prepared material for N-glycopeptides from standard protein digest samples. Moreover, it was further applied to simultaneous capturing of N- and O-glycopeptides from mouse liver protein digests. Compared to the commercially available zwitterionic hydrophilic interaction liquid chromatography (ZIC-HILIC) material, the number of glycoproteins corresponding to all N- and O-glycopeptides enriched with PDA-ADE@KCP was much more than that with ZIC-HILIC. Furthermore, PDA-ADE@KCP captured more O-glycopeptides than ZIC-HILIC, revealing its superior performance in O-glycopeptide enrichment. All these results indicated that the strategy holds immense potential in characterizing N- and O-intact glycopeptides in the field of proteomics.

Development of targeted hydrophilic interaction liquid chromatography-tandem mass spectrometry method for acyl-Coenzyme A covering short- to long-chain species in a single analytical run

Acyl-CoAs play a significant role in numerous physiological and metabolic processes making it important to assess their concentration levels for evaluating metabolic health. Considering the important role of acyl-CoAs, it is crucial to develop an analytical method that can analyze these compounds. Due to the structural variations of acyl-CoAs, multiple analytical methods are often required for comprehensive analysis of these compounds, which increases complexity and the analysis time. In this study, we have developed a method using a zwitterionic HILIC column that enables the coverage of free CoA and short- to long-chain acyl-CoA species in one analytical run. Initially, we developed the method using an LC-QTOF instrument for the identification of acyl-CoA species and optimizing their chromatography. Later, a targeted HILIC-MS/MS method was created in scheduled multiple reaction monitoring mode using a QTRAP MS detector. The performance of the method was evaluated based on various parameters such as linearity, precision, recovery and matrix effect. This method was applied to identify the difference in acyl-CoA profiles in HepG2 cells cultured in different conditions. Our findings revealed an increase in levels of acetyl-CoA, medium- and long-chain acyl-CoA while a decrease in the profiles of free CoA in the starved state, indicating a clear alteration in the fatty acid oxidation process.

Determination of 27 bovine plasma amino acids and metabolites using zwitterionic-hydrophilic interaction liquid chromatography coupled with isotope dilution electrospray ionization triple quadrupole liquid chromatography-mass spectrometry and the effect of deproteinization timing

The use of zwitterionic-hydrophilic interaction liquid chromatography (Z-HILIC) columns for analysis of underivatized analytes has allowed simpler sample preparation of bovine plasma for sensitive and selective analysis, when coupled with mass spectrometry (MS). The objective of this study was to evaluate and validate this analytical technique to measure AA and metabolites in bovine plasma at 2 deproteinization times. A robust method using Z-HILIC coupled to a triple quadrupole MS (TQMS) was evaluated and validated to quantitatively analyze 19 AA using isotope dilution and 8 AA metabolites qualitatively in bovine deproteinized plasma. The timing of deproteinization was investigated to determine if plasma should be deproteinized upon collection (on-site) or immediately before analysis (in-lab). Analytes were separated using a Z-HILIC column in a 21 min run and analyzed with a TQMS in positive electrospray ionization for identification and quantification. The method was validated for standard curve linearity, limits of detection (LOD) and quantification (LOQ), intra- and interday precision (% coefficient of variation; CV), recovery (%), and freeze-thaw stability (% CV) after 1 mo. Coefficients of determination (R2) were over 0.993, and LOD and LOQ were below measured values for all AA. The CV for the intraday and interday precision were below 18%, except for cystine (Cys2) and Orn in-lab. Recoveries on-site and in-lab ranged from 75% to 120% for all analytes except Cys2 in-lab. Most analytes were stable after 1 mo of freezing regardless of deproteinization timing, CV <25%, except for hydroxyproline (Hyp). The concentration of Cys2 was affected by deproteinization in-lab compared with on-site, and even though Glu and Hyp were different between the 2 deproteinization timings, the concentrations between the 2 timings were within the standard deviation.

A Single-Run HPLC-MS Multiplex Assay for Therapeutic Drug Monitoring of Relevant First- and Second-Line Antibiotics in the Treatment of Drug-Resistant Tuberculosis.

The treatment of drug-resistant Mycobacterium tuberculosis relies on complex antibiotic therapy. Inadequate antibiotic exposure can lead to treatment failure, acquired drug resistance, and an increased risk of adverse events. Therapeutic drug monitoring (TDM) can be used to optimize the antibiotic exposure. Therefore, we aimed to develop a single-run multiplex assay using high-performance liquid chromatography-mass spectrometry (HPLC-MS) for TDM of patients with multidrug-resistant, pre-extensively drug-resistant and extensively drug-resistant tuberculosis. A target profile for sufficient performance, based on the intended clinical application, was established and the assay was developed accordingly. Antibiotics were analyzed on a zwitterionic hydrophilic interaction liquid chromatography column and a triple quadrupole mass spectrometer using stable isotope-labeled internal standards. The assay was sufficiently sensitive to monitor drug concentrations over five half-lives for rifampicin, rifabutin, levofloxacin, moxifloxacin, bedaquiline, linezolid, clofazimine, terizidone/cycloserine, ethambutol, delamanid, pyrazinamide, meropenem, prothionamide, and para-amino salicylic acid (PAS). Accuracy and precision were sufficient to support clinical decision making (≤±15% in clinical samples and ±20-25% in spiked samples, with 80% of future measured concentrations predicted to fall within ±40% of nominal concentrations). The method was applied in the TDM of two patients with complex drug-resistant tuberculosis. All relevant antibiotics from their regimens could be quantified and high-dose therapy was initiated, followed by microbiological conversion. In conclusion, we developed a multiplex assay that enables TDM of the relevant first- and second-line anti-tuberculosis medicines in a single run and was able to show its applicability in TDM of two drug-resistant tuberculosis patients.

Comparison of ZIC-HILIC Columns for the Simultaneous Analysis of Antiviral Drugs in Dosage Forms by Hydrophilic Interaction Liquid Chromatography

Background:: Antiviral drugs are vital since many viruses can produce fatal infections, as we have recently seen with the COVID-19 pandemic. Antiviral drugs can battle viruses at multiple stages of their life cycles, including neuraminidase, nucleic acid synthesis, protease, and virion fusion or entry [1]. Objective:: Antiviral drugs have poor retention in reversed-phase liquid chromatography, which makes it difficult to analyze a mixture of antiviral medications using high-performance liquid chromatography. Using zwitterionic hydrophilic interaction liquid chromatography (ZICHILIC), the paper highlights the simultaneous quantification of three antiviral drugs as active constituents in pharmaceutical formulations. Moreover, the influence of the length of the spacer between the two charges in two stationary phases on the retention behavior of antiviral drugs has been discussed. Methods:: Two homemade stationary phases (ZIC1-HILIC and ZIC5-HILIC) were utilized for the qualitative and quantitative analysis of three antiviral drugs, and UV was used as the detector. Several chromatographic conditions were examined, such as the organic modifier concentration, buffer concentration, and pH value. Results:: After optimizing the parameters, the devised method was applied to analyse three antiviral medications quantitatively. The initial results demonstrated the current procedure for separating and determining these three antiviral drugs to be sensitive, robust, and effective. Consequently, the present method has shown excellent repeatability, a broad linear range (0.1- 16.5 μgml-1), and excellent sensitivity (LOD 0.04-0.072 μgml-1). The RSD value of the method was less than 1. Conclusion:: A mixed mode of hydrophilic and ion exchange interactions was the predominant mode of antiviral medications with two ZIC-HILIC stationary phases. The ZIC5-HILIC stationary phase had a lower detection and limit of quantitation for three antiviral drugs and a prolonged retention time compared to the ZIC1-HILIC stationary phase with a shorter chain length.

In-Depth Performance Analysis and Comparison of Monolithic and Particulate Zwitterionic Hydrophilic Interaction Liquid Chromatography Polymer Columns.

The kinetic performance of different zwitterionic hydrophilic interaction liquid chromatography polymer columns is evaluated and compared in-depth. For this purpose, two lab-made monolithic columns, synthesized with different crosslinkers, and a commercial particle packed column are considered. It is found that performance evaluation techniques, such as comparing plate height curves or fitted A-, B- and C-terms, obtained by fitting experimental plate height data to a plate height model, are complicated by the determination of a reliable characteristic length. This is due to the very different morphology of these column types, and the heterogeneity of the monolithic columns. The occurrence of a convective flow through the packed particle column further complicates the interpretation of the obtained fitting parameters, as part of the C-term is wrongfully attributed to the A-term. Therefore, the use of the kinetic plot method is suggested for the comparative evaluation of these columns, as kinetic plots do not require the determination of a characteristic length, nor rely on any fitting parameters. With the kinetic plot method, it is demonstrated that the lab-made monolithic columns outperform the packed particle column for plate counts between 10,000 and 800,000. This is attributed to the higher column efficiency of these columns, due to their small domain and skeleton sizes, and their high permeability, resulting from their high external porosity and the occasional occurrence of preferential flow paths.

The evaluation of Two Zwitterionic Hydrophilic Interaction Liquid Chromatography Materials for the Rapid Separation of Methamphetamine and Muscimol Pharmaceuticals

The investigation aimed to use two Zwitterionic Hydrophilic Interaction Liquid Chromatography (ZIC-HILIC) columns to examine the retention behavior of methamphetamine and muscimol. The various spacer lengths between zwitterionic molecules in the stationary phases have been utilized to investigate methamphetamine and muscimol retention behavior. In hydrophilic interaction liquid chromatography (HILIC), the retention characteristic of these two drugs was examined using a mixture of sodium acetate buffer/acetonitrile as an eluent equipped with a UV detector. It has been established that hydrophilic and hydrophobic mechanisms primarily govern the separation of methamphetamine and muscimol medicines on two ZIC-HILIC columns. As previously stated, the ZIC-HILIC-1 column demonstrated a difference when compared to the ZIC-HILIC-5 columns. The combination of positively and negatively charged locations in a single molecule bonded to the surface of an adsorbent allows for distinct variations in methamphetamine and muscimol selectivity separation.

Strategies for the Separation and Quantification of Non-Steroidal Anti- Inflammatory Drugs Using ZIC-HILIC-HPLC with UV Detection

Background: Non-steroidal anti-inflammatory drugs (NSAIDs) are widely used worldwide for their analgesic, anti-inflammatory, and antipyretic properties. Objective: The article highlights the development of zwitterionic hydrophilic interaction liquid chromatography (ZIC-HILIC) methods to quantify three NSAIDs as active ingredients in pharmaceutical formulations simultaneously. Method: For NSAIDs analysis, two homemade ZIC-HILIC stationary phases were carried out by adding sulfobetaine monomers to polystyrene-divinylbenzene particles. The methods were developed by examining the effect of HILIC mode parameters like the kind of organic modifier (methanol or acetonitrile), acetonitrile content, pH and concentration of the acetate buffer in the eluent. Results: The experimental data exhibit the predominant mechanism of NSAIDs with two ZICHILIC stationary phases. The different chain lengths in the two ZIC-HILIC stationary phases are employed to investigate NSAIDs retention behaviour. The strategies were evaluated for their specificity, linearity, accuracy, and precision. The linear ranges were 0.01-5.0, 0.05-17.5 and 0.12-13.5 μg ml-1, LOD 0.005. 0.015 and 0.046 μg ml-1 and LOQ 0.015, 0.045 and 0.139 μg ml-1 for nimesulide, tenoxicam and diclofenac, respectively. Conclusion: They represent interactions with three NSAIDs, ZIC-HILIC columns, and mobile phase in a mixed-mode of hydrophilic and hydrophobic interactions. In contrast to the ZICHILIC- 1 stationary phase with a shorter chain length, the ZIC-HILIC-4 stationary phase with a longer chain length exhibited a longer retention time, lower detection and limit of quantitation for three NSAIDs.The verification findings demonstrate the suitability of the proposed ZIC-HILIC methods for quantifying NSAIDs in pharmaceutical formulations.

Analytical quality by design approach for the determination of imidazole in sildenafil API and its formulations using zwitterionic HILIC stationary phase

Herein, the development of a HILIC method for the determination of imidazole (Imp E) in sildenafil citrate API and its final formulations is reported. The main goal of this study was to develop a robust, application-specific HPLC method according to the Analytical Quality by Design principles for the analysis of the above impurity. After the risk assessment study, the high-risk method parameters were sequentially screened and optimized by using 2-level fractional factorial and Box-Behnken designs. The mathematical models were combined with the Monte-Carlo simulations to identify the Method Operable Design Region. The method was thoroughly validated between 25 % and 150 % of the target concentration limit of the imidazole using the total-error concept. The relative bias varied between 1.6 % and 5.6 % and the RSD values were lower than 5.8 % for repeatability and intermediate precision. The limit of detection and the lower limit of quantification were satisfactory and found to be 0.025 and 0.125 μg mL−1 imidazole, respectively. The applicability of the proposed approach has been demonstrated in the analysis of several sildenafil citrate API batches and final products.

On the occurrence of very low intra-particle diffusion rates in zwitterionic hydrophilic interaction liquid chromatography polymer columns

Intra-particle diffusion is investigated in particle-packed and monolithic zwitterionic hydrophilic interaction liquid chromatography (HILIC) polymer columns. For this purpose, plate heights are first measured over a broad range of velocities for polar compounds with zone retention factors between k″ = 2 and 10 on all column types, multiplied with the corresponding velocity, and extrapolated to zero velocity. The thus obtained B-term coefficients are additionally verified via peak parking experiments and indicate a very low degree of longitudinal diffusion in all columns, that moreover seems to be independent of the zone retention factor. This is in contrast to earlier observations on reversed-phase liquid chromatography columns and bare silica HILIC columns. To obtain more insight into the reasons for these low levels of longitudinal diffusion, the experimentally obtained effective diffusion coefficients are modeled to equations derived from the effective medium theory, such as the models developed by Torquato and Jefrey & Hashin. It is, however, demonstrated that these models fail to adequately describe effective diffusion in the studied zwitterionic HILIC columns, due to the low degree of intraparticle diffusion in these stationary phases. Fitting the experimental effective diffusion coefficients to the two limiting cases of the effective medium theory, i.e., the pure parallel-connection and pure serial-connection case, it is observed that to stay within these limits, intraparticle diffusion must be extremely low, i.e., Dpz/Dm ≤ 0.01, indicating that the diffusion inside the pores is at least 100 times smaller than the bulk diffusion coefficient. There are several possible explanations for these low intraparticle diffusion coefficients, such as a strongly hindered diffusion in the polymer matrix, extremely low surface diffusion rates in the aqueous layer adsorbed onto the stationary phase and/or the occurrence of slow localized adsorption events.

Methods for Quantifying the Metabolic Boundary Fluxes of Cell Cultures in Large Cohorts by High-Resolution Hydrophilic Liquid Chromatography Mass Spectrometry.

Metabolomics is a mainstream approach for investigating the metabolic underpinnings of complex biological phenomena and is increasingly being applied to large-scale studies involving hundreds or thousands of samples. Although metabolomics methods are robust in smaller-scale studies, they can be challenging to apply to larger cohorts due to the inherent variability of liquid chromatography mass spectrometry (LC-MS). Much of this difficulty results from the time-dependent changes in the LC-MS system, which affects both the qualitative and quantitative performances of the instrument. Herein, we introduce an analytical strategy for addressing this problem in large-scale microbial studies. Our approach quantifies microbial boundary fluxes using two zwitterionic hydrophilic interaction liquid chromatography (ZIC-HILIC) columns that are plumbed to enable offline column equilibration. Using this strategy, we show that over 397 common metabolites can be resolved in 4.5 min per sample and that metabolites can be quantified with a median coefficient of variation of 0.127 across 1100 technical replicates. We illustrate the utility of this strategy via an analysis of 960 strains of Staphylococcus aureus isolated from bloodstream infections. These data capture the diversity of metabolic phenotypes observed in clinical isolates and provide an example of how large-scale investigations can leverage our novel analytical strategy.

Solute–Solvent Interactions in Hydrophilic Interaction Liquid Chromatography: Characterization of the Retention in a Silica Column by the Abraham Linear Free Energy Relationship Model

The Abraham linear free energy relationship model has been used to characterize a hydrophilic interaction liquid chromatography (HILIC) silica column with acetonitrile/water and methanol/water mobile phases. Analysis by the model for acetonitrile/water mobile phases points to solute volume and hydrogen bond basicity as the main properties affecting retention, whereas solute hydrogen bond acidity, dipolarity and polarizability practically do not affect it. Formation of a cavity is easier in acetonitrile-rich mobile phases than in the aqueous stationary phase, and hence increase of solute volume decreases retention. Conversely, hydrogen bond acidity is stronger in the aqueous stationary phase than in the acetonitrile-rich mobile phase and thus an increase of solute hydrogen bond basicity increases retention. Results are similar for methanol/water mobile phases with the difference that solute hydrogen bond acidity is significant too. Increase in hydrogen bond acidity of the solute decreases retention showing that methanol mobile phases must be better hydrogen bond acceptors than acetonitrile ones, and even than water-rich stationary phases. The results are like the ones obtained in zwitterionic HILIC columns bonded to silica or polymer supports for acetonitrile/water mobile phases, but different for solute hydrogen bond acidity for a polymer bonded zwitterionic column with methanol/water mobile phases, indicating that bonding support plays an important role in HILIC retention. Comparison to RPLC characterized systems confirms the complementarity of HILIC systems to RPLC ones because the main properties affecting retention are the same but with reversed coefficients. The least retained solutes in RPLC are the most retained in HILIC.

Application of a hybrid zwitterionic hydrophilic interaction liquid chromatography column in metabolic profiling studies

Metabolic phenotyping studies using mouse liver extracts as a model, performed on a novel zwitterionic HILIC UHPLC column, which is based on ethylene-bridged hybrid organic/inorganic particles bonded with sulfobetaine groups and packed into column hardware modified with hybrid surface technology are reported. Initially the chromatographic performance was evaluated under different mobile phase conditions using selected metabolite standards. Following optimization of the chromatographic conditions for 88 hydrophilic metabolites both targeted and untargeted profiling analyses were performed on tissue extracts using LC-MS/MS and LC-TOF/MS, respectively.Chromatographic efficiency parameters such as peak resolution, peak shapes, selectivity and precision in retention and peak areas as well as characteristics that are critical for metabolic profiling analysis such as metabolite coverage and retention time distribution were assessed.The hybrid zwitterionic column exhibited efficient chromatographic separations providing analysis of ca 80 hydrophilic metabolites from different chemical classes and polarities. Utilizing a one-dimensional separation both targeted and untargeted profiling provided comprehensive metabolic signatures that enabled the acquisition of the metabolic phenotypes of the tissue extracts.

One-step preparation of magnetic zwitterionic–hydrophilic dual functional nanospheres for in-depth glycopeptides analysis in Alzheimer's disease patients' serum

A novel ultra-hydrophilic zwitterionic-HILIC (ZIC-HILIC) nanosphere (Fe3O4-CG) was synthesized via a one-step hydrothermal strategy, which significantly simplified the conventional multi-step procedures for the preparation of ZIC-HILIC materials. The dual-functional Fe3O4-CG nanosphere exhibited excellent selectivity (molar ratio BSA:HRP = 5000:1), low detection limit (0.05 fmol/μL), satisfactory reusability (at least 5 times) and recovery rate (93.7 ± 2.1%). The binding constant of Fe3O4-CG for HRP is 2.45 ± 0.32 × 10−6 M and the theoretical binding capacity is 330 mg g−1. In addition, the Fe3O4-CG microsphere showed excellent performance in the detection of glycopeptides from real biological samples. Furthermore, 131 glycopeptides related to 71 glycoproteins were selectively enriched from healthy human serum and 180 glycopeptides related to 82 glycoproteins were captured from Alzheimer's disease patients' serum analyzed by Nano-LC-MS/MS. Gene ontology analysis of the biological process and molecular function showed that 21 primitive glycoproteins in glycopeptides captured from Alzheimer's disease patients' serum were meaningfully involved in a variety of neurodegenerative disease-related events, including serine-type endopeptidase inhibitor activity, receptor binding, positive regulation of B cell activation, and platelet activation.

Monitoring of Paraquat in soya products intended for animal feed

Even though paraquat is considered to be one of the most toxic herbicides, it is one of the most used ones. Paraquat-based preparations are heavily used across many important agricultural countries and for a lot of types of crops such as soya. As soya is used in large quantities in the feed production, paraquat residues in soya products are a potential threat to livestock. The aim of this study was to investigate whether soya products intended for animal feed contain residues of paraquat. Therefore, an optimized Quick Polar Pesticides (QuPPe) Method was developed. In total, 174 samples were analyzed via LC-MS/MS using zwitterionic hydrophilic interaction liquid chromatography (ZIC-HILIC). The results showed that paraquat was detected in 66 % of the samples above 0.008 mg/kg. Residues were mainly present in soya bean meals that were imported from Brazil or the USA and cultivated using genetically modified soya. Around 5 % of the samples were not compliant with the European regulation on maximum residue levels of pesticides in or on food and feed. The results of this study support the view that paraquat residues play a significant role in soya products intended for animal feed and, thus, need to be controlled on a regular basis. However, for enforcement actions reliable processing factors for paraquat in soya products or maximum residue levels for processed soya products are urgently required.

Analytical Problems in Separation of Selenomethionine and Its Oxidative Product in HILIC HPLC.

Selenomethionine (SeMet) is one of the main selenium forms in foods and supplements. Determining its presence in natural food samples creates difficulties due to possible oxidation processes. The objective of this study was to evaluate the possible degradation of SeMet in water extracts of green teas, one of the most consumed beverages worldwide. Such a medium has not been investigated at this time. The HILIC-HPLC MS/MS method with different stationary phases was used to achieve the satisfactory separation of SeMet and selenomethionine oxide (SeMetO). The addition of dithiothreitol and β-mercaptoethanol, recommended to ensure that SeMet is kept in the reduced form, was also evaluated. The best separation was achieved using the zwitterionic HILIC stationary phase coupled to mass spectrometry and MeOH with water (85/15, v/v) as the eluent. Extraction was done with hot water with the addition of β-mercaptoethanol. The infusions prepared from Lung-Ching teas (from the Zhejiang Province in China) contained the highest concentration of selenium in a typical cup of tea (12.5–17.3 µg L−1). For other tested teas it decreased in the following order: Yunnan > Dilmah > Lipton. For Lung-Ching teas, the sum of concentrations of SeMet and SeMetO corresponded to about 46–63% of the total selenium in their extracts.

Characterization and Separation of Platinum-Based Antineoplastic Drugs by Zwitterionic Hydrophilic Interaction Liquid Chromatography (HILIC)–Tandem Mass Spectrometry, and Its Application in Surface Wipe Sampling

Platinum-based antineoplastic drugs (PtADs) are among the most important and used families of chemotherapy drugs, which, even showing severe side effects and being hindered by drug resistance, are not likely to be replaced clinically any time soon. The growing interest in the occupational health community in antineoplastic drug (AD) surface contamination requires the development of increasingly fast and easy high-throughput monitoring methods, even considering the lack of harmonized legally binding regulation criteria. Thus, a wipe sampling method together with zwitterionic hydrophilic interaction liquid chromatography (HILIC-Z)–tandem mass spectrometry (MS/MS) analysis was developed for the simultaneous evaluation of oxaliplatin, cisplatin, and carboplatin surface contaminations. A design of experiments approach was used to optimize the chromatographic conditions. Limits of quantification ranging from 2 to 5 ng/mL were obtained from interday and intraday repetitions for oxaliplatin and carboplatin, and between 170 and 240 ng/mL for cisplatin. The wipe desorption procedure is equivalent to other AD sampling methods, enabling a fast sample preparation, with an LC-MS/MS analysis time of less than 7 min.
